RNA干扰(RNAi)是真核生物中普遍存在的一种自然现象,利用双链RNA诱发宿主细胞同源mRNA降解,从而抑制特异目的基因的表达,不仅具有理论意义,而且具有实用价值。白血病是造血系统常见的恶性肿瘤,虽然通过联合化疗大多数患者可以获得完全缓解,但是最终不可避免的是多药耐药(MDR)导致化疗的失败。MDR已成为目前治愈白血病的最大障碍。因此,逆转MDR成为白血病治疗亟待解决的问题。本文以白血病MDR机制的阐述为线索,综述了RNAi技术逆转白血病MDR的研究进展。
RNA interference (RNAi) is a ubiquitous natural phenomenon in eukaryotes. It is not only of theoretical significance but also of practical value to use double-stranded RNA to induce homologous mRNA degradation in host cells to inhibit the expression of a specific target gene. Leukemia is a common malignant tumor of the hematopoietic system. Although complete remission can be achieved in most patients with combination chemotherapy, ultimately it is unavoidable that multidrug resistance (MDR) results in the failure of chemotherapy. MDR has become the biggest obstacle to cure leukemia. Therefore, the reversal of MDR has become an urgent problem to be solved in the treatment of leukemia. In this paper, the exposition of MDR mechanism of leukemia as a clue, reviewed RNAi technology to reverse the progress of leukemia MDR research.
