This paper reports the use of periodic acid-linked avidin and horseradish peroxidase for ELISA to increase the sensitivity of standard ELISA for the detection of human IgG. The ratio of avidin to activated horseradish peroxidase is related to the sensitivity and the ratio of 1: 1.5 is the most sensitive. The chemical modification of avidin - horseradish enzyme and avidin - horseradish enzyme were compared. The former was used in BA-ELISA, the sensitivity limit was 4 times higher than that of standard ELISA, and the experimental results were quite stable. Avidin-HRP was used in BA-ELISA, which was 4-16 times sensitive than normal ELISA. However, Non-specific adsorption and high coefficient of variation are not acceptable for most uses. Other experimental factors that may improve BA-ELISA are discussed.
