对怀地黄试管苗的脱毒技术进行了研究.结果发现:以MS为基本培养基并附加适宜浓度细胞分裂素(6-BA0.5 mg.L-1或KT0.5 mg.L-1)时,茎尖成苗率最高(75%),出愈率较低(20%);切离的茎尖大小为0.2~0.5 mm时,既能保证较高的脱毒率又可保证较高的成苗率;两次切离茎尖并与热处理相结合,试管苗的脱毒率最高(96%~100%),建立了一种有效的怀地黄脱毒方法和技术.
The technology of detoxification of the radix rehmanniae in vitro was studied.It was found that MS with basal medium supplemented with appropriate concentration of cytokinin (6-BA0.5 mg.L-1 or KT0.5 mg.L-1) (75%), the rate of emergence was lower (20%). When the tip size of cuttings was 0.2-0.5 mm, the higher rate of detoxification and higher Of seedling rate; twice cut off the shoot tip and combined with heat treatment, the highest rate of virus-free in vitro (96% ~ 100%), established an effective detoxification methods and techniques of Rehmannia.
