目的了解转录因子GATA3基因在白血病和正常骨髓基质细胞(BMSC)中表达的情况。方法体外扩增培养BMSC,运用RTPCRELISA方法检测GATA3基因的表达情况,并对其相对表达水平进行半定量比较。结果GATA3基因在正常和白血病的BMSC中均有一定的表达。BMSC中急性髓性白血病(AML)和慢性粒细胞白血病(CML)组的GATA3基因表达率与正常组比较无明显差异,而急性淋巴细胞白血病(ALL)组的GATA3表达率比正常组低,且AML的BMSC中GATA3基因表达水平>ALL>正常>CML。结论转录因子GATA3基因可在正常和白血病骨髓基质细胞中表达,并可能影响骨髓微环境的造血调控作用,其表达模式的改变可能与白血病发生发展有一定的关系。
Objective To investigate the expression of transcription factor GATA3 gene in leukemia and normal bone marrow stromal cells (BMSC). Methods BMSCs were expanded and cultured in vitro. The expression of GATA3 gene was detected by RTPCRELISA, and the relative expression levels of GATA3 were compared semi-quantitatively. Results The GATA3 gene was expressed in normal and leukemic BMSCs. There was no significant difference of GATA3 gene expression between AML and BML in BMSC compared with that of normal group, but GATA3 expression in ALL group was lower than that of normal group AML BMSC GATA3 gene expression levels> ALL> normal> CML. Conclusion The transcription factor GATA3 gene can be expressed in normal and leukemia bone marrow stromal cells and may affect hematopoietic regulation of bone marrow microenvironment. The change of its expression pattern may be related to the occurrence and development of leukemia.
