猪胆汁中胆酸皂化工艺研究

胆酸是存在于动物胆汁中,经皂化分离提取的一种生化物质,化学名称是3α,7α-二羟基-5β-胆烷酸。研究采用胆红素和胆酸联产工艺方案,以胆汁提取胆红素生产过程中的滤去胆钙盐的母液为原料,选取HCl浓度、pH值、NaOH浓度、皂化时间和料液比5个因素进行单因素实验,进而选取NaOH浓度、皂化时间和料液比3个因素进行皂化工艺正交实验。研究表明,HCl浓度1∶2、pH值3.0、NaOH浓度6%、皂化时间12h、料液比1∶10为猪胆汁中总胆酸最佳皂化工艺条件。 Cholic acid is a biochemical substance found in animal bile that has been extracted by saponification and has the chemical name 3α, 7α-dihydroxy-5β-cholanoic acid. The study adopted bilirubin and bile acid co-production process, bile extract bilirubin production process to filter out the bile calcium salt mother liquor as raw material, select the HCl concentration, pH, NaOH concentration, saponification time and the ratio of 5 Factor single factor experiments, and then choose the NaOH concentration, saponification time and the ratio of material to liquid saponification process orthogonal experiment. The results showed that the optimum conditions for the total saponification of total cholic acid in pig bile were HCl concentration 1: 2, pH value 3.0, NaOH concentration 6%, saponification time 12h and solid-liquid ratio 1:10.
