目的观察1~6型蛋白精氨酸甲基转移酶(PRMTs)在E3大鼠急性哮喘模型肺脏和脾脏中的表达变化。方法构建卵清蛋白诱导的E3大鼠急性哮喘模型,分为对照组和哮喘组,每组各10只大鼠,病理及生化检测评估模型构建情况,半定量PCR检测1~6型PRMTs的表达差异。结果和对照组相比,哮喘组大鼠肺脏组织中PRMT1(P<0.01)、PRMT2(P<0.01)、PRMT3(P<0.05)、PRMT5(P<0.05)的表达有显著性升高,而PRMT4(P<0.05)的表达显著性降低;哮喘组大鼠脾脏组织中PRMT2(P<0.05)、PRMT5(P<0.05)的表达也出现显著性升高。结论蛋白精氨酸甲基转移酶在E3大鼠急性哮喘模型肺脏和脾脏中有差异性表达,这可能在哮喘发病的基因转录后调节过程中发挥着重要的作用。
Objective To investigate the expression changes of type 1 - 6 protein arginine methyltransferase (PRMTs) in the lung and spleen of acute asthmatic model of E3 rats. Methods The acute asthma model induced by ovalbumin in E3 rats was established and divided into control group and asthma group with 10 rats in each group. The pathological and biochemical tests were used to evaluate the model construction. The expression of PRMTs in type 1 to 6 was detected by semi-quantitative PCR difference. Results Compared with the control group, the expression of PRMT1 (P <0.01), PRMT2 (P <0.01), PRMT3 (P <0.05) and PRMT5 (P <0.05) The expression of PRMT4 (P <0.05) decreased significantly in asthmatic rats. The expression of PRMT2 (P <0.05) and PRMT5 (P <0.05) in asthmatic rats also increased significantly. Conclusions Protein arginine methyltransferase is differentially expressed in the lung and spleen of the acute asthmatic model of E3 rats, which may play an important role in the post-transcriptional regulation of asthma pathogenesis.
