目的:探讨TXNDC9基因在结肠直肠癌组织和细胞中的表达,以及沉默TXNDC9高表达后对细胞株SW1116增殖、迁移、侵袭能力的影响。方法:分别应用免疫组织化学技术、免疫印迹法观察TXNDC9在结肠直肠癌组织和细胞中的表达水平:应用小干扰RNA瞬时转染TXNDC9高表达的结肠癌细胞株SW1116,用实时PCR筛选最高效率的干扰片段,并用Western印迹法检测基因沉默效果:最后研究瞬时转染后SW1116细胞增殖、迁移、侵袭能力的变化。结果:结肠直肠癌组织中TXNDC9的表达明显高于对照的癌旁正常组织(P<0.01),Western印迹法证实TXNDC9在SW1116、SW620、SW480、Colo205四株细胞中呈高表达,在HCT116和DLD1细胞中表达最少。在成功沉默SW1116细胞中TXNDC9基因的表达后,与两对照组相比,干扰组在72~96 h内细胞增值能力显著下降(P<0.01),细胞迁移和侵袭能力亦有明显下降(P<0.01)。结论:TXNDC9在结肠直肠癌组织中的表达显著高于癌旁正常黏膜上皮,并与肿瘤的大小和浸润深度相关,SW1116是TXNDC9基因高表达的细胞株之一,下调TXNDC9表达能显著抑制该细胞的增殖能力、细胞迁移和侵袭能力。TXNDC9基因对维持肿瘤细胞的增殖和运动能力有重要作用,并可能成为抑制肿瘤生长和转移的有效途径。
Objective: To investigate the expression of TXNDC9 in colorectal cancer tissues and cells and the effect of TXNDC9 silencing on proliferation, migration and invasion of SW1116 cells. Methods: The expressions of TXNDC9 in colorectal cancer tissues and cells were detected by immunohistochemistry and Western blotting respectively: Transfection of TXNDC9-overexpressing colon cancer cell line SW1116 with small interfering RNA was performed. Real-time PCR was used to screen the highest efficiency The fragment was silenced and the gene silencing effect was detected by Western blotting. Finally, the changes of proliferation, migration and invasion of SW1116 cells after transient transfection were studied. Results: The expression of TXNDC9 in colorectal cancer tissues was significantly higher than that in the normal tissues (P <0.01). Western blotting confirmed that TXNDC9 was highly expressed in four cell lines of SW1116, SW620, SW480 and Colo205. In HCT116 and DLD1 The least expressed in cells. After successfully silenced the expression of TXNDC9 gene in SW1116 cells, the proliferation ability of SW1116 cells was significantly decreased (P <0.01) and the migration and invasion ability of SW1116 cells was also significantly decreased from 72 h to 96 h (P < 0.01). CONCLUSION: The expression of TXNDC9 in colorectal cancer tissues is significantly higher than that in adjacent normal mucosal epithelium, and is related to the size and depth of invasion. SW1116 is one of the cell lines with high expression of TXNDC9 gene. Downregulation of TXNDC9 expression can significantly inhibit this cell Proliferation ability, cell migration and invasion ability. The TXNDC9 gene plays an important role in maintaining the proliferation and motility of tumor cells and may be an effective way to inhibit tumor growth and metastasis.
