目的研究益肺解毒方联合顺铂诱导人肺腺癌A549细胞凋亡的增效机制。方法应用血清药理学方法制备益肺解毒方药物血清,然后采用高剂量含药血清与顺铂联合作用于人肺腺癌A549细胞,同时设置对照组、空白血清组、高剂量药物血清组、顺铂组进行对照试验,48h后应用免疫细胞化学方法进行Bax、Bcl-2表达水平测定,对比分析不同分组方式对细胞不同效果的凋亡作用。结果与空白对照组比较,空白血清对人肺腺癌A549细胞Bax和Bcl-2表达无统计学差异(P>0.05),高剂量药物血清、顺铂、高剂量益肺解毒方协同顺铂组均具有促进人肺腺癌A549细胞Bax表达的作用(P<0.05),但高剂量药物血清与顺铂协同后对人肺腺癌Bax表达促进作用最为明显。同时还发现,高剂量药物血清、顺铂、高剂量益肺解毒方协同顺铂具有抑制人肺腺癌A549细胞Bcl-2表达的作用,同样高剂量药物血清与顺铂协同后对人肺腺癌Bcl-2表达抑制作用更明显。结论益肺解毒方与顺铂协同增效机制可能是通过促进Bax基因的表达、抑制Bcl-2基因的表达,从而诱导肿瘤细胞凋亡。
Objective To study the synergistic mechanism of Yifeng Jiedu decoction combined with cisplatin on apoptosis of human lung adenocarcinoma A549 cells. Conclusion The synergistic mechanism of Yifu Jiedu Recipe and cisplatin may induce apoptosis of tumor cells by promoting the expression of Bax gene and inhibiting the expression of Bcl-2 gene.
