目的:制备临床分离的多重耐药大肠埃希菌TH1、TH2、TH6、TH7和TH10的脂多糖(LPS)脂质体并观察其对肿瘤的抑制作用。方法:采用逆向蒸发法制备大肠埃希菌LPS脂质体,通过电镜、粒径测定仪,鲎试验法(LAL)测定LPS脂质体的包封率。LPS脂质体免疫小鼠后将单肿瘤细胞悬液分别接种于小鼠腹腔和腓肠肌内,观察LPS脂质体对腹水瘤和实体肿瘤的作用。结果:大肠埃希菌LPS脂质体均为乳白色悬液,镜下可见LPS脂质体均呈规则的球形,粒径分布均匀。LAL结果推算包封率为99.0%~99.9%。TH1、TH2、TH6、TH7和TH10LPS脂质体免疫小鼠与对照组相比,腹水量和细胞总数明显减少且体质量增加,P<0.05,小鼠腓肠肌瘤体的体积均小于对照组,P<0.05。结论:所制备的大肠埃希菌LPS脂质体对肿瘤具有抑制作用。
OBJECTIVE: To prepare lipopolysaccharide (LPS) liposomes of clinically isolated multidrug-resistant Escherichia coli TH1, TH2, TH6, TH7 and TH10 and to observe their inhibitory effect on tumor. Methods: LPS liposomes of Escherichia coli were prepared by reverse evaporation. The encapsulation efficiency of LPS liposomes was determined by electron microscopy, particle size analyzer and LAL assay. LPS liposomes were immunized mice single tumor cell suspension were inoculated into the abdominal cavity and gastrocnemius mice to observe the role of lipopolysaccharide on ascites tumor and solid tumors. Results: The LPS liposomes of Escherichia coli were all milky white suspension. The LPS liposomes showed regular spherical shape with uniform particle size distribution. LAL results estimated encapsulation efficiency of 99.0% ~ 99.9%. TH1, TH2, TH6, TH7 and TH10LPS liposome immunized mice compared with the control group, ascites and total cell number decreased significantly and body weight increased, P <0.05, the size of the mice gastrocnemius tumor were smaller than the control group, P <0.05. Conclusion: The prepared LPS liposome of Escherichia coli can inhibit the tumor.
