目的:建立离子色谱法测定谷胱甘肽等含酰胺基药物中游离铵盐的含量。方法:采用IonPac CS12A色谱柱(250mm×4mm,8μm),IonPac CG12A预柱(50mm×4mm,8μm),以18mmol·L-1甲烷磺酸溶液为淋洗液,流速1.0mL·min-1,电导检测器检测。结果:NH4+在0.203~4.07μg·mL-1浓度范围内有良好的线性关系,r=0.9995;最低检测限为0.025ng;平均加样回收率在92.1%~100.6%之间。结论:本法简便、快速、准确,灵敏度高,重复性好,适用于含酰胺基药物中游离铵盐的质量控制。
OBJECTIVE: To establish a method for the determination of free ammonium salt in amido-containing drugs such as glutathione by ion chromatography. Methods: IonPac CS12A column (250mm × 4mm, 8μm) and IonPac CG12A precolumn (50mm × 4mm, 8μm) were used in this experiment. The flow rate was 1.0mL · min-1 with 18mmol·L- Conductivity detector detection. Results: The linear range of NH4 + was 0.203 ~ 4.07μg · mL-1, r = 0.9995. The minimum detectable limit was 0.025ng. The average recovery was between 92.1% and 100.6%. Conclusion: The method is simple, rapid, accurate, sensitive and reproducible. It is suitable for quality control of free ammonium salts in amido-containing drugs.
