目的 :观察中药复方复聪片及拆方后方内补肾、益气养血、活血祛瘀、通窍聪耳等主要药物对豚鼠庆大霉素 (GM )耳毒性反应的拮抗作用。方法 :豚鼠每日肌肉注射GM80mg/kg,同时每日喂服复聪片或等效剂量各拆方组药物 ,连续 2 0日 ,测定用药前后听觉脑干诱发电位 (ABR)阈值 ,耳蜗基底膜铺片外毛细胞计数 ,螺旋韧带铺片观察血管纹血流状况及耳蜗超微结构电镜观察等 ,评价药物的拮抗效果及对耳蜗结构的保护作用。结果 :复聪片及其拆方之补肾、益气养血、活血祛瘀药物可减轻GM所致耳蜗外毛细胞损伤率 (P <0 0 5 ) ,保护耳蜗结构及功能 ,拮抗GM所致之听阈升高 (P <0 0 5 )。结论 :复聪片及方中补肾、益气养血、活血祛瘀药物可有效减轻GM的耳毒性反应。
Objective : To observe the antagonism of the ototoxicity response of gentamicin (GM ) in guinea pigs with traditional Chinese medicine compound Fusong tablets and disassembled prescriptions of main drugs such as kidney-tonifying, Qi-nourishing, blood-activating, and Tongqiao-Chai. METHODS: Guinea pigs received daily intramuscular injection of GM80mg/kg, and were fed daily with Fusong tablets or equivalent doses in each group. On the 20th consecutive day, the threshold of auditory brain stem evoked potential (ABR) was measured before and after administration. The basement membrane of the cochlea was measured. The number of outer hair cells was counted, the spiral ligament spread was used to observe the blood flow of stria vascularis, and the ultrastructure of the cochlea was observed by electron microscopy. The antagonism of the drug and the protective effect on the cochlea structure were evaluated. Results: Fusong tablets and their decomposed prescriptions of kidney-tonifying, Qi-nourishing, and blood-activating drugs can reduce the damage rate of GM-induced outer hair cells (P < 0.05), protect the structure and function of the cochlea, and antagonize GM. The hearing threshold increased (P < 0 05). Conclusion : Fusong tablets and Fangzhongzhong Bushen, Nourishing Qi and activating blood stasis drugs can effectively reduce the ototoxicity response of GM.
