目的 :研究机械牵张应变对人牙周膜细胞(human periodontal ligament cells,HPDLCs)晚期凋亡的影响。方法 :体外培养HPDLCs,利用动态机械应变细胞加载装置对HPDLCs加载1%、10%和20%的动态牵张应变6、12、24和48 h,通过流式细胞仪和激光扫描共聚焦显微镜对经过TUNEL染色的细胞进行晚期凋亡检测。采用SPSS 11.0软件包对数据进行统计学分析。结果:HPDLCs加载后,呈现栅栏状相互平行排列趋势,排列方向垂直于牵张力方向。各应变组细胞的晚期凋亡率在加载24 h时达到该应变组的峰值,且具有一定时间和应变依赖性。结论:通过动态机械应变细胞加载装置对人牙周膜细胞的牵张加载,发现牵张应变可以诱导细胞发生晚期凋亡,具有时间和应变值依赖性,24 h时晚期凋亡率达到峰值。
Objective: To investigate the effect of mechanical stretch on the late apoptosis of human periodontal ligament cells (HPDLCs). METHODS: HPDLCs were cultured in vitro. HPDLCs were loaded with 1%, 10% and 20% dynamic strain at dynamic stress-strain for 6, 12, 24 and 48 h respectively by dynamic mechanical strain cell loading device. Flow cytometry and laser scanning confocal microscopy TUNEL-stained cells were tested for late apoptosis. Data were analyzed statistically using SPSS 11.0 software package. Results: After loaded, the HPDLCs showed a palisade-like arrangement parallel to each other with the arrangement direction perpendicular to the direction of strain. The late apoptotic rate of each strain group reaches the peak of this strain group at 24 h after loading, and has a certain time and strain-dependent. CONCLUSION: Stretching and loading of human periodontal ligament cells by dynamic mechanical strain cell loading device shows that stretch-induced strain can induce late apoptosis, which is time-dependent and strain-dependent. The late apoptosis rate reaches its peak at 24 hours.
