目的 探讨大鼠胃黏膜细胞EGFR活化AP -1的信号传导通路。方法 用EGFR配体TGF -α刺激分离的大鼠胃黏膜细胞 ,Westernblot和EMSA方法检测ERK -1/ 2信号传导通路的活化程度。结果 1nmol/LTGF -α刺激胃黏膜细胞 3 0min不仅显著诱导AP -1的活性 ,而且明显激活MEK和ERK -1/ 2 ;EGFR特异性抑制剂PD15 3 0 3 5或MEK特异性抑制剂PD980 5 9能完全阻断TGF -α诱导的胃黏膜细胞AP -1活化。结论 大鼠胃黏膜细胞EGFR通过ERK -1/ 2信号传导通路激活AP -1。
Objective To investigate the signal transduction pathway of EGF-activated AP-1 in rat gastric mucosal cells. Methods The isolated rat gastric mucosal cells were stimulated with EGFR ligand TGF-α, and the activation of ERK-1/2 signal transduction pathway was detected by Western blot and EMSA. Results The gastric mucosal cells treated with 1nmol / L of TGF-α for 30min not only significantly induced AP-1 activity but also significantly activated MEK and ERK-1/2; EGFR-specific inhibitor PD15 3 0 3 5 or MEK-specific inhibitor PD980 5 9 completely blocked TGF-α-induced AP-1 activation in gastric mucosal cells. Conclusion EGFR can activate AP-1 through the ERK-1/2 signaling pathway in rat gastric mucosa.
