△6-脂肪酸脱氢酶基因可将亚油酸转化为γ-亚麻酸。我们将克隆的深黄被孢霉△6-脂肪酸脱氢酶基因(D6D)插入植物表达载体pBI121,构建了重组质粒pBI121-D6DMI。通过农杆菌(Agrobacterium tumefaciens)介导途径,将深黄被孢霉△6-脂肪酸脱氢酶基因导入了甘蓝型油菜恢复系,经过诱导分化获得抗卡那霉素的再生植株。PCR检测及Souther杂交结果表明,外源基因△6-脂肪酸脱氢酶基因已整合到油菜再生植株的基因组中。
The △ 6-fatty acid dehydrogenase gene converts linoleic acid to γ-linolenic acid. We cloned the gene of C.fumigatus △ 6-fatty acid dehydrogenase (D6D) into the plant expression vector pBI121 to construct the recombinant plasmid pBI121-D6DMI. The Agrobacterium tumefaciens-mediated pathway was used to transfer the △ 6-fatty acid desaturase gene of Mortierella wasabi to the restorer line of Brassica napus, and the regenerated plants resistant to kanamycin were obtained through induction and differentiation. PCR results and Souther hybridization results showed that the exogenous gene △ 6-fatty acid dehydrogenase gene has been integrated into the genome of rapeseed regenerated plants.
