【目的】初步探讨白藜芦醇(resveratrol,Res)抗白血病的免疫学机制。【方法】用MTT比色法测定肿瘤细胞生长抑制率,流式细胞术分析其细胞周期。分别采用溶血分光光度法、改良的Mayer法及双抗体夹心ELISA法,检测荷瘤小鼠的免疫功能及细胞因子含量变化。【结果】在一定范围内Res可以时间-剂量依赖的方式抑制白血病L1210细胞增殖、促进其凋亡。Res可促进荷瘤小鼠T细胞增殖、抗体的生成,并且上调IL-2、减少IL-8的分泌水平。【结论】Res可直接抑制白血病L1210细胞增殖,并且通过调节机体免疫应答的水平,协同发挥抗肿瘤作用。
【Objective】 To investigate the immunological mechanism of resveratrol (Res) against leukemia. 【Methods】 The growth inhibition rate of tumor cells was determined by MTT colorimetric method, and the cell cycle was analyzed by flow cytometry. The immune function and cytokine content in tumor-bearing mice were detected by hemolysis spectrophotometry, modified Mayer method and double antibody sandwich ELISA respectively. 【Results】 Res inhibited the proliferation and promoted the apoptosis of leukemic L1210 cells in a time-and dose-dependent manner. Res can promote T cell proliferation, antibody production in tumor-bearing mice, and up-regulate IL-2 and decrease the secretion of IL-8. 【Conclusion】 Res can directly inhibit the proliferation of leukemia L1210 cells and synergistically exert antitumor effects by regulating the level of immune response.
