目的探讨聚合人脐带血血红蛋白(PolyPHb)对新西兰大耳白兔心肌缺血/再灌注损伤的保护效果。方法采用随机分组对照研究的方法,将30只雄性新西兰大耳白兔随机平均分为假手术组、对照组和实验组;对照组和实验组分别注射5 ml生理盐水和PolyPHb溶液(8 gHb/dl)做预处理,通过临时阻断冠脉前降支实现心肌缺血;观察缺血30 min、再灌注90 min后的心脏功能和肌钙蛋白Ⅰ释放水平。结果实验组与对照组比较,兔左室收缩压:(82.59±8.13)mmHg vs(61.39±7.92)mmHg(P<0.05);左室内压最大上升速率:(3.76±0.73)mmHg/msvs(2.23±0.89)mmHg/ms(P<0.05);左室内压最大下降速率:(-3.08±0.49)mmHg/ms vs(-1.89±0.65)mm-Hg/ms,P<0.05):左室舒张末压(9.35±1.82)mmHg vs(18.21±2.43)mmHg(P<0.01);肌钙蛋白Ⅰ释放水平:(8.71±1.90)ng/ml vs(17.83±1.72)ng/ml(P<0.05)。结论 PolyPHb预处理对兔心肌缺血/再灌注损伤具有明显保护作用。
Objective To investigate the protective effect of PolyHb on myocardial ischemia / reperfusion injury in New Zealand white rabbits. Methods 30 male New Zealand white rabbits were randomly divided into sham operation group, control group and experimental group. The control group and experimental group were injected with 5 ml normal saline and PolyPHb solution (8 gHb / dl) for preconditioning. Myocardial ischemia was achieved by temporarily blocking the anterior descending coronary artery. The cardiac function and troponin I release were observed at 30 min after ischemia and 90 min after reperfusion. Results Compared with the control group, the left ventricular systolic pressure in experimental group was (82.59 ± 8.13) mmHg vs (61.39 ± 7.92) mmHg (P <0.05), and the maximum rate of left ventricular pressure was (3.76 ± 0.73) mmHg / msvs ± 0.89) mmHg / ms (P <0.05); The maximum rate of decrease of left ventricular pressure was (-3.88 ± 0.49) mmHg / ms vs (-1.89 ± 0.65) mmHg / (9.35 ± 1.82) mmHg vs (18.21 ± 2.43) mmHg (P <0.01). The release of troponin Ⅰ was (8.71 ± 1.90) ng / ml vs (17.83 ± 1.72) ng / ml, respectively. Conclusion PolyPHb preconditioning has significant protective effect on myocardial ischemia / reperfusion injury in rabbits.
