目的通过同位素示踪技术探讨氟尿嘧啶脂质体(FL)的局部淋巴结靶向性,为恶性肿瘤的局部淋巴靶向化疗提供依据。方法将6只兔双侧第一趾蹼皮下注射99mTc-FL后,用SPECT连续动态示踪30min或60min,然后在8h内静态显像监测FL的淋巴流向途径,并探测1、3、6h时在注射区、窝淋巴结、盆腔淋巴结群、后腹膜淋巴结群、乳糜池等局部淋巴结区域的闪烁计数率(%)。结果动态示踪、静态监测见注射区99mTc-FL缓慢靶向流至局部淋巴结而显影,局部淋巴结高浓度积聚,而其余组织浓度很低。在注药后1、3、6h3组间注射区或局部淋巴结的闪烁计数率比较,差异有统计学意义(P<0.05),注射区的闪烁计数率逐渐下降,而局部淋巴结则逐渐上升;各组内局部淋巴结与其余组织(注射区除外)的闪烁计数率比较差异有统计学意义(P<0.05),局部淋巴结的闪烁计数率明显高于其余组织。结论FL具有局部淋巴靶向性和富集性,并不直接进入血液循环,提示恶性肿瘤局部淋巴靶向化疗具有可行性。
Objective To investigate the target localization of fluorouracil liposomes (FL) by isotope tracer and to provide a basis for local lymph targeted chemotherapy in malignant tumors. Methods 99mTc-FL was injected subcutaneously into the bilateral first toe fins of 6 rabbits, followed by SPECT continuous dynamic tracing for 30min or 60min, and then statically observed within 8h to monitor the lymphatic flow pathways of FL and to detect 1,3,6h The scintillation counting rate (%) in the regional lymph nodes in the injection zone, popliteal lymph node, pelvic lymph node group, retroperitoneal lymph node group, chylomicrons. The results of dynamic tracing, static monitoring of injection area 99mTc-FL slow targeting flow to the local lymph nodes and imaging, local lymph node accumulation of high concentrations, while the rest of the tissue concentration is low. The scintillation counting rate of injection area or local lymph node between 1, 3, 6h3 groups after injection was significantly different (P <0.05), scintigraphy rate of injection area was gradually decreased and local lymph node was gradually increased. There was significant difference in scintillation counting rate between local lymph nodes and other tissues (except for injection area) (P <0.05). The scintigraphic count rate of local lymph nodes was significantly higher than that of other tissues. Conclusion FL has local lymph targeting and enrichment, does not directly enter the blood circulation, suggesting that local lymph node-targeted chemotherapy of malignant tumor is feasible.
