通过探讨一种新的钙依赖性磷脂结合蛋白Annexin32对家兔凝血时间和兔、大鼠血栓形成的影响,对其主要药效学进行了研究,并与普通肝素进行了比较.兔、大鼠静脉给药,检测用药后不同时间家兔白陶土部分凝血活酶时间(KPTT)、体外血栓形成和大鼠体内血栓形成的变化.结果显示,按1mg/kg静脉注射Annexin32后,KPTT明显延长(P<0.01),但小于肝素延长幅度,体外血栓形成仪测定显示血栓长度、重量均明显减少(P<0.01),大鼠体内血栓重量明显降低(P<0.01),提示Annexin32具有抗血栓形成的功能,并有对凝血系统影响小的特征.
By studying the effect of Annexin32, a new calcium-dependent phospholipid binding protein, on the clotting time of rabbits and the thrombosis in rabbits and rats, the main pharmacodynamics were studied and compared with that of unfractionated heparin (KPTT), thrombosis in vitro and thrombus formation in rats were measured at different time points after administration.Results showed that KPTT was significantly prolonged after 1 mg / kg intravenous injection of Annexin32 (P <0.01), but less than the prolongation of heparin. The thrombus measured in vitro showed that the length and weight of thrombus decreased significantly (P <0.01), and the weight of thrombus decreased significantly in rats (P <0.01), suggesting that Annexin32 has antithrombotic Function, and have little effect on the coagulation system features.
