目的 观察冠心胶囊对大鼠心肌梗死的影响及其作用机制。方法 用大鼠冠脉结扎法制备心肌梗死模型 ,应用膜片钳技术研究药物对L 型钙通道的作用。结果 冠心胶囊能剂量依赖性地对抗大鼠冠脉结扎引起的心肌梗死 ,并明显抑制心肌缺血引起的S T段抬高。冠心胶囊 1,10 ,30mg·ml-1可显著抑制钙通道 ,减少钙离子内流 ,30mg·ml-1可抑制钙离子 5 2 % (P <0 .0 1)。结论 冠心胶囊能剂量依赖性地缩小大鼠心肌梗死面积 ,对心梗具有保护作用。这一作用可能和其调节植物神经的功能 ,抑制钙通道 ,稳定心肌细胞膜有关。
Objective To observe the effect and mechanism of Guanxin Capsule on myocardial infarction in rats. Methods Myocardial infarction model was established by rat coronary artery ligation and patch clamp technique was used to study the effect of drugs on L-type calcium channels. Results Guanxin capsule can dose-dependently treat myocardial infarction caused by coronary artery ligation in rats, and significantly inhibit ST elevation in myocardial ischemia. Guanxin Capsules 1, 10 and 30 mg·ml-1 significantly inhibited calcium channels and reduced calcium influx. 30 mg·ml-1 inhibited calcium ion 5 2% (P < 0.01). Conclusion Guanxin Capsule can reduce the myocardial infarct size of rats in a dose-dependent manner and has a protective effect on myocardial infarction. This effect may be related to its function of regulating the autonomic nervous system, inhibiting calcium channels and stabilizing the myocardial cell membrane.
