该研究采用家兔内毒素血症模型 ,探讨磷脂酶A2 (PLA2 )抑制剂磷酸氯喹 (CQ)对内毒素血症时细胞、亚细胞膜的保护效应。实验结果表明 ,注射内毒素后MAP显著下降 ,8h存活率为 5 7% ;血浆、红细胞膜、肝线粒体膜PLA2 活性及MDA含量显著升高 ,并随着伤情的加重而升高。而CQ预处理内毒素组注射内毒素后MAP下降幅度显著小于内毒素组 (P <0 .0 5 ) ,血浆、红细胞膜、肝线粒体膜PLA2 活性及MDA含量也显著低于内毒素组 ,动物 8h活存率达 87.5 %。PLA2 抑制剂CQ能大大提高动物活存率和活存时间 ,改善血液动力学 ,抑制PLA2 活性 ,保护细胞膜。提示 :CQ在抗休克和全身炎症方面有较好效果
In this study, rabbit endotoxemia model was used to investigate the protective effect of phospholipase A2 (CQ2) on cells and subcellular membranes during endotoxemia. The results showed that MAP significantly decreased after injection of endotoxin, and the survival rate was 57% at 8h. PLA2 activity and MDA content in plasma, erythrocyte membrane and liver mitochondrial membrane were significantly increased, and increased with the severity of injury. However, the decrease of MAP in CQ pretreated LPS group was significantly lower than that of LPS group (P <0.05), and the PLA2 activity and MDA content in plasma, erythrocyte membrane and liver mitochondrial membrane were also significantly lower than those in LPS group 8h survival rate of 87.5%. PLA2 inhibitor CQ can greatly improve animal survival rate and survival time, improve hemodynamics, inhibit PLA2 activity, protect the cell membrane. Tip: CQ in anti-shock and systemic inflammation have a good effect
