目的对转化生长因子TGF-β与结缔组织生长因子CTGF在石英导致的肺纤维化过程中的作用进行综述。方法通过查阅国内外与TGF-β、CTGF以及石英致肺纤维化相关的文献资料,分析TGF-β、CTGF在石英所致肺纤维化过程中作用。结果石英暴露可导致TGF-β升高,而TGF-β又通过Smad依赖途径和Smad非依赖途径诱导CTGF的生成。TGF-β启动肺纤维化,并通过产生高水平的CTGF来进一步维持持续的纤维化反应,而CTGF表达增加可能才是石英导致肺纤维化发生发展的中心环节。结论 CTGF在TGF-β的调控下参与了石英所致肺部纤维化的发展与维持。
Objective To review the role of transforming growth factor (TGF-β) and connective tissue growth factor (CTGF) in the process of quartz-induced pulmonary fibrosis. Methods By referring to literature data related to TGF-β, CTGF and quartz induced pulmonary fibrosis at home and abroad, the role of TGF-β and CTGF in quartz-induced pulmonary fibrosis was analyzed. RESULTS: Quartz exposure resulted in elevated TGF-β, which in turn induced CTGF production through Smad-dependent and Smad-independent pathways. TGF-beta initiates pulmonary fibrosis and further maintains sustained fibrotic response by producing high levels of CTGF, whereas increased expression of CTGF may be central to the development of quartz-induced pulmonary fibrosis. Conclusion CTGF is involved in the development and maintenance of pulmonary fibrosis induced by quartz under the regulation of TGF-β.
