目的 研究定位于 12 q2 3的核心蛋白聚糖 (decorin)基因与高度近视的关系。方法 收集高度近视先证者 90例 ,制备外周血白细胞基因组 DNA,应用 PCR法分别扩增核心蛋白聚糖基因 8个外显子及其邻近内含子 ,然后用异源双链 - SSCP法分析 PCR产物 ,寻找可能的变异。结果 分析 90例高度近视先证者核心蛋白聚糖基因全部 8个外显子及其邻近内含自子 ,未发现任何变异。结论 核心蛋白聚糖基因突变可能与本组高度近视无关。decorin基因与人类疾病的关系有待进一步研究
Objective To study the relationship between the decorin gene localized to 12q2 3 and high myopia. Methods Totally 90 cases of high myopia proband were collected to prepare genomic DNA of peripheral blood leukocytes. Eight exon of decorin gene and its adjacent introns were amplified by PCR, and then analyzed by heteroduplex - SSCP PCR products, looking for possible variations. RESULTS: All eight exons of the decorin gene in 90 cases of high myopia probands and their neighboring introns contained no variations. Conclusion The decorin gene mutation may not be related to this group of high myopia. The relationship between decorin gene and human diseases needs further study
