背景:糖尿病心肌病的发病机制尚不清楚,神经调节蛋白1具有促进血管再生等作用,糖尿病心肌病的发病是否和心肌微血管内皮细胞表达神经调节蛋白1下降有关值得进一步研究。目的:观察高糖对心肌微血管内皮细胞表达神经调节蛋白1的影响。方法:取生长良好的第2代心肌微血管内皮细胞,高糖组加入10mmol/L的葡萄糖,高糖+胰岛素组加入10mmol/L的葡萄糖及10-5U/L的胰岛素,对照组正常培养。培养24h后收集细胞,RT-PCR及Westernblot方法检测神经调节蛋白1mRNA和蛋白的表达。结果与结论:与对照组比较,高糖组心肌微血管内皮细胞神经调节蛋白1mRNA和蛋白的表达明显下降(P<0.05);与高糖组比较,高糖+胰岛素组心肌微血管内皮细胞神经调节蛋白1mRNA和蛋白的表达明显升高(P<0.05),与对照组间差异无显著性意义(P>0.05)。说明高糖可抑制心肌微血管内皮细胞表达神经调节蛋白1mRNA和蛋白,而胰岛素可拮抗此作用。
BACKGROUND: The pathogenesis of diabetic cardiomyopathy is unclear. Neuregulin 1 plays a role in promoting blood vessel regeneration. Whether diabetic cardiomyopathy is associated with decreased expression of neuregulin 1 in myocardial microvascular endothelial cells deserves further study. Objective: To observe the effect of high glucose on the expression of neuregulin 1 in myocardial microvascular endothelial cells. Methods: The second generation of myocardial microvascular endothelial cells with good growth were obtained. Glucose of 10mmol / L and glucose of 10mmol / L and insulin of 10-5U / L were added into the high glucose and insulin groups. The control group was cultured normally. The cells were collected after culturing for 24h, and the expression of neuregulin 1 protein and protein were detected by RT-PCR and Western blot. RESULTS AND CONCLUSION: Compared with the control group, the expression of neuregulin 1 mRNA and protein in myocardial microvascular endothelial cells was significantly decreased in high glucose group (P <0.05). Compared with high glucose group, the expression of neuregulin 1 mRNA and protein were significantly increased (P <0.05), but there was no significant difference between the two groups (P> 0.05). High glucose can inhibit the expression of neuregulin 1 mRNA and protein in myocardial microvascular endothelial cells, and insulin can antagonize this effect.
