目的:探讨测定组织中基质金属蛋白酶(MMPs)的方法,了解MMPs活性的变化与分娩发动的关系。方法:利用生物素标记的明胶作底物,与组织中MMPs作用后,用酶联免疫法测定剩余明胶的含量,据此测出酶的活性。结果:以此法测定了分娩不同阶段的子宫下段和宫颈中MMPs活性,发现总MMPs和基质金属蛋白酶-9(MMP-9)的活性,从未临产、分娩潜伏期、分娩活跃期、宫口开全依次升高,各组之间相差显著。结论:基质金属蛋白酶参与了分娩的发动;生物素标记的明胶可用于测定组织中MMPs活性。
OBJECTIVE: To investigate the method of determination of matrix metalloproteinases (MMPs) in tissues and to understand the relationship between changes of MMPs activity and labor onset. Methods: By using biotin-labeled gelatin as substrate and the effect of MMPs in the tissue, the content of the remaining gelatin was determined by enzyme-linked immunosorbent assay, and the enzyme activity was measured. Results: The activity of MMPs and matrix metalloproteinase-9 (MMP-9) in the lower uterine segment and cervix at different stages of labor were measured by this method. The results showed that the activity of MMPs and MMP- All followed by increased, a significant difference between the groups. CONCLUSIONS: Matrix metalloproteinases are involved in the onset of labor; biotinylated gelatin can be used to determine MMPs activity in tissues.
