目的研究洛泊对体外培养的浆液性卵巢癌细胞SKOV3凋亡作用及探讨其对凋亡基因bcl-2和bax表达的影响。方法体外培养卵巢癌细胞SKOV3,用MTT比色法检测洛铂对SKOV3细胞株的增殖抑制作用以及流式细胞仪检测洛铂对SKOV3细胞株的凋亡率,流式细胞仪检测凋亡相关蛋白bax、bcl-2的表达。结果体外培养卵巢癌SKOV3细胞经不同浓度洛泊处理后细胞生长明显受到抑制,洛泊作用于卵巢癌SKOV3细胞可诱导细胞的凋亡,同时,随着洛铂浓度的增加,bax基因的表达增加,而bcl-2表达减少。结论卵巢癌SKOV3细胞对洛泊具有药物敏感性,洛泊可诱导卵巢癌细胞的凋亡,bax蛋白的表达增加和bcl-2蛋白表达的减少可能是洛泊诱导卵巢癌细胞的凋亡机制之一。
Objective To investigate the apoptosis of ovarian carcinoma cell line SKOV3 induced by losanopril and to explore its effect on the expression of bcl-2 and bax genes. Methods The ovarian cancer cell line SKOV3 was cultured in vitro. The inhibitory effect of lobaplatin on SKOV3 cell line was detected by MTT colorimetric assay. The apoptosis rate of SKOV3 cell line was detected by flow cytometry. The apoptosis-related protein bax, bcl-2 expression. Results The growth of ovarian cancer cell line SKOV3 was inhibited by different doses of oxaliplatin in vitro. The effect of opal administration on ovarian cancer cell SKOV3 induced cell apoptosis. At the same time, the expression of bax gene was increased with the increase of lobaplatin concentration , While the bcl-2 expression decreased. CONCLUSION: Ovarian cancer SKOV3 cells are drug sensitive to lobesorb and can induce the apoptosis of ovarian cancer cells. The increase of bax protein and the decrease of bcl-2 protein expression may be the mechanism of the apoptosis of ovarian cancer cell line OPOVA one.
