三七总皂甙对甲基硝基亚硝基胍转化的人胃黏膜上皮细胞GES-1的促凋亡作用

目的 从细胞分子水平探讨活血化瘀中药三七治疗胃癌前病变的作用机制。方法 以甲基硝基亚硝基胍 (MNNG)转化人胃黏膜上皮细胞系GES 1细胞 ,采用电镜、活细胞荧光染色和末端脱氧核苷酸转移酶标记法 (TUNEL法 ) ,检测三七总皂甙 (PNS)作用 4 8h对转化细胞 (MC细胞 )的促凋亡作用 ,并以维甲酸 (RA)作为对照。结果  2 0 0mg/LPNS作用 4 8h有显著的促进转化细胞凋亡的作用 (与MC组比较 ,P <0 .0 1) ,而 10 -5mol/LRA则对MC细胞凋亡比例无明显影响 (P >0 .0 5 )。结论 三七有促进MNNG转化的人胃黏膜上皮细胞凋亡的作用 ,这可能是三七治疗胃癌前病变的作用机制之一。 Objective To explore the mechanism of traditional Chinese medicines for promoting blood circulation and removing blood stasis on precancerous lesions of gastric cancer from the molecular level of cells. METHODS: Human gastric epithelial cell line GES 1 cells were transformed with MNNG and subjected to electron microscopy, live cell fluorescence staining and terminal deoxynucleotidyl transferase labeling (TUNEL assay). The effect of saponin (PNS) on the apoptosis of transformed cells (MC cells) for 48 hours and retinoic acid (RA) as a control. RESULTS: The effect of 200 mg/LPNS for 48 h had significant effect on apoptosis of transformed cells (P < 0.01 compared with MC group), while 10 -5 mol/LRA had no significant effect on MC apoptosis rate (P <0.01). P >0 .0 5 ). Conclusion Panax notoginseng has the effect of promoting the apoptosis of gastric epithelial cells in MNNG transformation, which may be one of the mechanisms of Panax notoginseng treatment of gastric precancerous lesions.
