目的探讨水中草颗粒对复发性口腔溃疡细胞免疫功能的影响。方法采用免疫方法建立复发性口腔溃疡动物模型,将成模的复发性口腔溃疡家兔随机分为模型组、左旋咪唑组(2.5mg/mL)、水中草低剂量组(0.25g/mL)和高剂量组(0.5g/mL),同时设立正常组。从实验的第7周开始,左旋咪唑组、水中草低剂量组及水中草高剂量组每天分别以相应药物灌胃,共连续28天;取耳缘静脉血检测T淋巴细胞亚群(CD3、CD4、CD8)、血清白细胞介素(IL)-10和IL-12的水平。结果模型组家兔CD3、CD4及CD4/CD8比值[分别为(40.50±7.46)%、(30.80±5.33)%及79.56±8.32]均较正常组显著下降(P<0.05),血清IL-10水平[(8.02±0.53)ng/L]较正常组升高(P<0.05),IL-12水平[(15.51±1.35)ng/L]较正常组下降(P<0.05);血清CD3、CD4、CD4/CD8比值及IL-12水平,左旋咪唑[(55.30±6.33)%、(43.50±5.33)%、99.29±13.84及(17.72±1.70)ng/L]、水中草低剂量[(58.50±6.03)%、(42.80±3.17)%、110.27±12.85及(17.74±1.96)ng/L]、水中草高剂量组[(61.50±4.32)%、(47.20±4.37)%、103.30±7.42及(18.18±1.54)ng/L]均较模型组升高(P<0.05,P<0.01),血清IL-10水平左旋咪唑组、水中草低剂量组和水中草高剂量组均较模型组降低(P<0.05,P<0.01)。结论复发性口腔溃疡家兔免疫功能处于抑制状态,水中草颗粒能够改善复发性口腔溃疡家兔的免疫抑制状态,达到治疗复发性口腔溃疡的作用。
Objective To investigate the effects of Suo Cao Granule on cellular immune function of recurrent oral ulcers. Methods The animal model of recurrent oral ulcer was established by immunization method. The model of recurrent oral ulcer rabbits was randomly divided into model group, levamisole group (2.5 mg/mL), indigo grass low-dose group (0.25 g/mL) and high. Dosage group (0.5g/mL), while the normal group was established. From the 7th week of the experiment, the levamisole group, the low dose group of the aquatic herb, and the high dose group of the aquatic herb were fed with the corresponding drugs for 28 consecutive days. The ear venous blood was used to detect the T lymphocyte subsets (CD3, CD4, CD8), serum interleukin (IL)-10 and IL-12 levels. Results The ratios of CD3, CD4 and CD4/CD8 in the model group were significantly lower than those in the normal group (40.50±7.46)%, (30.80±5.33)%, and 79.56±8.32, respectively (P<0.05). The level of [(8.02±0.53) ng/L] was higher than that of the normal group (P<0.05), and the IL-12 level [(15.51±1.35) ng/L] was lower than that of the normal group (P<0.05); the serum CD3, CD4 , CD4/CD8 ratios and IL-12 levels, levamisole [(55.30±6.33)%, (43.50±5.33)%, 99.29±13.84 and (17.72±1.70) ng/L], low dose of aquatic grass [(58.50± 6.03)%, (42.80±3.17)%, 110.27±12.85 and (17.74±1.96) ng/L], high dose of aquatic grasses [(61.50±4.32)%, (47.20±4.37)%, 103.30±7.42 and ( 18.18±1.54) ng/L] were higher than those in the model group (P<0.05, P<0.01). Serum IL-10 levels were lower in the levamisole group, the lower dose group of the aquatic herb, and the high dose group of the aquatic herb than the model group ( P<0.05, P<0.01). Conclusion The immune function of recurrent oral ulcers is inhibited. Shuixue Caojing can improve the immunosuppressive state of recurrent oral ulcers in rabbits and achieve the effect of recurrent oral ulcers.
