目的:研究神经干细胞冻存复苏后的成活率及再培养生长情况。方法:利用无血清培养基短期体外培养胚胎来源的神经干细胞后,以冻存液(体积分数为70% DMEM/F12,体积分数为20%胎牛血清,10% DMSO)进行冻存,于1周、4周、8周、12周、16周、20周、24周分别复苏,计数活细胞比例并进行再培养。结果:复苏后神经干细胞的成活比例分别为68%、65%、65%、62%、61%、60%、60%,再培养生长良好。结论:本实验对神经干细胞成功地进行了冻存、复苏和再培养,对临床择期进行神经干细胞移植手术和建立“神经干细胞库”具有重要的意义。
Objective: To study the survival rate and the growth of neural stem cells after cryopreservation and resuscitation. Methods: Embryonic neural stem cells were cultured in serum-free medium for a short period of time and then cryopreserved in a cryopreservation medium (volume fraction of 70% DMEM / F12, 20% fetal calf serum, 10% DMSO) Weeks, 4 weeks, 8 weeks, 12 weeks, 16 weeks, 20 weeks, 24 weeks were resumed, the proportion of living cells were counted and re-cultured. Results: The survival rate of neural stem cells after resuscitation were 68%, 65%, 65%, 62%, 61%, 60% and 60%, respectively. Conclusion: The successful cryopreservation, resuscitation and re-culturing of neural stem cells in this experiment is of great significance for the clinical elective neural stem cell transplantation and the establishment of neural stem cell bank.
