目的 建立 4月龄人胎肝组织差异表达基因EST库 ,并分离未知功能基因以寻找造血相关的新基因。方法 表达性差异显示技术 (RDA)、核苷酸序列分析、竞争PCR及Northern杂交。结果 建立了 4月龄人胎肝组织差异表达基因EST库。该文库富集了造血及肝生长调控相关基因 ,部分克隆序列测定及分析表明该库含有重要的未知功能新基因 ,具有一株新基因可能与红细胞增殖、分化有关。结论 RDA技术为寻找新的基因提供了有效的手段。该EST库内富含了与造血相关基因包括一株新基因
OBJECTIVE: To establish a EST library of differentially expressed genes in 4-month-old human fetal liver tissue and to isolate unknown functional genes to find novel genes related to hematopoiesis. Methods Expression differential display technique (RDA), nucleotide sequence analysis, competitive PCR and Northern hybridization. Results The 4-month-old human fetal liver tissue EST library was established. The library is enriched in genes related to hematopoietic and hepatic growth regulation. The determination and analysis of some cloned sequences indicate that the library contains important new genes with unknown function. A new gene may be related to erythrocyte proliferation and differentiation. Conclusion RDA technology provides an effective means of finding new genes.
