目的:观察资木瓜总苷(TSCS)对佐剂性关节炎(AA)大鼠滑膜组织病理改变、滑膜肥大细胞脱颗粒及类胰蛋白酶表达的影响,探讨肥大细胞功能与TSCS治疗大鼠AA的关系。方法:雄性Wistar大鼠随机分为正常对照组、模型组和TSCS组(200 mg.kg-1),每组15只,致炎后第3天开始ig给药,1次/d,模型组和正常对照组给予等容积生理盐水,持续21 d。除正常对照组外,其余大鼠足部注射弗氏完全佐剂诱导建立AA模型,检测大鼠体质量和足跖体积变化,HE染色评价滑膜组织病理改变,TB染色观察滑膜肥大细胞数量及其脱颗粒情况,免疫组化法检测滑膜组织中类胰蛋白酶表达情况。结果:TSCS能明显增加AA大鼠体质量,减轻足跖肿胀度,显著减轻滑膜组织炎性细胞浸润、滑膜细胞增生和滑膜纤维组织增生,有效抑制滑膜肥大细胞数量和脱颗粒率,显著抑制滑膜组织类胰蛋白酶表达。相关分析显示,滑膜肥大细胞数量及脱颗粒率均与滑膜组织病理改变总评分呈明显的正相关。结论:TSCS具有治疗大鼠AA和调节滑膜肥大细胞功能的作用,由于两者存在明显的正相关性,因此TSCS可能通过抑制滑膜肥大细胞功能,对AA大鼠起治疗作用。
Objective: To observe the effect of TSCS on the pathological changes of synovial tissue, the degranulation of synovial mast cells and the expression of tryptase in adjuvant arthritis (AA) rats, and to explore the relationship between mast cell function and TSCS-treated rats AA’s relationship. Methods: Male Wistar rats were randomly divided into normal control group, model group and TSCS group (200 mg.kg-1), 15 rats in each group. And normal control group were given equal volume of saline for 21 days. In addition to the normal control group, other rats were injected with Freund’s complete adjuvant to establish AA model, the body weight and the plantar volume of the rats were measured, the pathological changes of synovial tissue were evaluated by HE staining, the number of synovial mast cells was observed by TB staining And its degranulation, immunohistochemistry of tryptase expression in synovial tissue. Results: TSCS significantly increased the body weight of AA rats, relieved the degree of paw edema, significantly reduced synovial infiltration of inflammatory cells, synovial cell proliferation and synovial fibrosis, and effectively inhibited the number of synovial mast cells and the rate of degranulation , Significantly inhibited synovial tissue tryptase expression. Correlation analysis showed that the number of synovial mast cells and degranulation rate were significantly positively correlated with the gross pathological changes of synovial tissue. CONCLUSION: TSCS has the effect of treating rat AA and regulating the function of synovial mast cells. TSCS may play a therapeutic role in AA rats by inhibiting the function of synovial mast cells due to the obvious positive correlation between them.
