目的 研究凋亡调控基因 bcl-2和 bax 在急性白血病(AL)中的表达、相互关系和临床意义。方法 应用半定量逆转录-聚合酶链反应(RT-PCR)和 Southern blot 技术检测32例 AL 患儿 bcl-2和 bax mRNA 水平及其比值。结果 AL 患儿骨髓细胞中 bcl-2 mRNA 的表达明显高于对照组(P<0.01),bax 的表达明显降低(P<0.01)。治疗后,bcl-2 mRNA 的表达在完全缓解(CR)组中明显低于未缓解(NR)组(P<0.01),但 bax mRNA 在两组间的表达无差异(P>0.05)。分析 bcl-2/bax mRNA 表达比值显示,高比值组 NR 率显著高于低比值组(P<0.01)。结论 AL 的发病与凋亡调控基因 bcl-2和 bax 的异常表达有关。bcl-2和 bax mRNA 及其比值可作为判断 AL 预后及指导治疗的主要监测指标。
Objective To study the expression, correlation and clinical significance of apoptosis-regulating genes bcl-2 and bax in acute leukemia (AL). Methods The mRNA and protein levels of bcl-2 and bax in 32 children with AL were detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and Southern blot. Results The expression of bcl-2 mRNA in bone marrow cells of children with AL was significantly higher than that of the control group (P <0.01), and the expression of bax was significantly decreased (P <0.01). After treatment, the expression of bcl-2 mRNA in CR group was significantly lower than that in untreated (NR) group (P <0.01), but there was no difference in bax mRNA expression between the two groups (P> 0.05). Analysis of bcl-2 / bax mRNA expression ratio showed that the high ratio group NR rate was significantly higher than the low ratio group (P <0.01). Conclusion The pathogenesis of AL is related to the abnormal expression of bcl-2 and bax genes. bcl-2 and bax mRNA and its ratio can be used as a predictor of AL prognosis and guide the treatment of the main indicators.
