目的 初步探讨临床乳腺癌细胞的细胞周期蛋白 (Cyclin)表达规律并予以归类 ,为进一步研究肿瘤的细胞周期机制提供依据。方法 采用Cyclin/DNA双参数流式细胞术对临床上获取的乳腺癌细胞的Cyclin (CyclinD1、E、A和B1)和DNA表达进行检测。结果 在 16例乳腺癌标本中 ,细胞周期蛋白的非时相性表达可以归类如下 :①Ⅰ型CyclinD1、E、A和B1均呈非时相性过表达 (5例 )。②Ⅱ型CyclinsD1、E、A和B1均呈低表达或不表达 (3例 )。③Ⅲ型CyclinD1呈过表达并贯穿于整个细胞周期 ,而CyclinsE、A和B1呈低表达或不表达 (3例 )。④Ⅳ型CyclinsE、A和B1呈不同程度的非时相性过表达 ,而CyclinD1呈低表达或不表达 (5例 )。结论 Cyclins在乳腺癌细胞中的非时相性表达存在着一定的规律 ,而每一种表达类型则显示了某一特定的细胞周期破坏机制
OBJECTIVE: To investigate and classify the expression of Cyclin in clinical breast cancer cells and to provide basis for further study on the cell cycle mechanism of tumor. Methods Cyclin (CyclinD1, E, A, B1) and DNA expression were detected by Cyclin / DNA dual-parameter flow cytometry in breast cancer cells. Results In 16 cases of breast cancer, the non-phase expression of cyclin can be classified as follows: ①Ⅰ, CyclinD1, E, A and B1 were non-phase-wise over-expression (5 cases). ② TypeⅡ CyclinsD1, E, A and B1 showed low or no expression (3 cases). ③ The type Ⅲ CyclinD1 was overexpressed and throughout the cell cycle, whereas CyclinsE, A and B1 showed low or no expression (3 cases). ④ The type Ⅳ CyclinsE, A and B1 showed different degrees of non-phase-over-expression, while CyclinD1 was low or no expression (5 cases). Conclusion There is a certain regularity of non-phase expression of Cyclins in breast cancer cells, and each type of expression shows a specific mechanism of cell cycle disruption
