目的探讨中度低温对内毒素性急性肺损伤(ALI)大鼠肺炎症反应的影响。方法采用大鼠内毒素性ALI模型,将34只雄性SD大鼠随机正常对照组(C组,8只);内毒素组(L组,10只)⑶低温组(H组,8只);内毒素+低温组(L+H组)。于ALI后4h处死大鼠,取肺组织行病理检查,并采用酶联免疫吸附法(ELISA)测定支气管肺泡灌洗液(BALF)中肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)和白介素-10(IL-10)含量。结果L组BALF中TNF-α、IL-6及IL-10含量较C组显著升高(P<0.05,P<0.01),而L+H组BALF中TNF-α、IL-6含量较L组显著下降(P<0.01),但两组间BALF中IL-10含量差异无显著性(P>0.05)。病理检查结果显示L+H组肺组织损伤减轻。结论32.5~33.0℃可减轻内毒素性ALI大鼠肺炎症反应。
Objective To investigate the effect of moderate hypothermia on the inflammatory reaction of lung in endotoxin-induced acute lung injury (ALI) rats. Methods Totally 34 male SD rats were randomly divided into normal control group (C group, n = 8), endotoxin group (group L, n = 10), hypothermia group (n = 8) Endotoxin + hypothermia group (L + H group). The rats were sacrificed at 4h after ALI and the lungs were removed for pathological examination. The levels of tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) in bronchoalveolar lavage fluid (BALF) were determined by enzyme-linked immunosorbent assay IL-6) and interleukin-10 (IL-10) content. Results The levels of TNF-α, IL-6 and IL-10 in BALF of L group were significantly higher than those of C group (P <0.05, P <0.01) (P <0.01). However, the level of IL-10 in BALF was not significantly different between the two groups (P> 0.05). Pathological examination showed that lung injury in L + H group was alleviated. Conclusion 32.5 ~ 33.0 ℃ can attenuate lung inflammation in endotoxic ALI rats.
