目的探讨建立适合骨髓基质干细胞移植于创面的动物模型,以研究骨髓基质干细胞在损伤修复中的作用机制。方法分离、培养、鉴定骨髓基质干细胞;在大鼠背部形成一4cm×4cm皮肤全层缺损创面;36只大鼠分3组(同种异体皮、脱细胞异种真皮、凡士林纱布)覆盖创面,将标记的P5代细胞移植于创面,观察覆盖物脱落时间、移植成活细胞数量。结果同种异体皮作为创面覆盖物比其他两组覆盖物脱落时间延长、成活细胞数量多;而脱细胞异种真皮覆盖物组又优于凡士林纱布覆盖物组。结论应用同种异体皮作为创面覆盖物能够较好地保护移植骨髓基质干细胞,成功构建一个创伤皮肤缺损模型用于研究骨髓基质干细胞的移植。
OBJECTIVE: To establish an animal model of bone marrow stromal stem cells transplanted to the wound in order to study the mechanism of bone marrow stromal stem cells in injury repair. METHODS: BMSCs were isolated, cultured, and identified. A 4 cm × 4 cm wound of full-thickness skin defect was formed on the back of rats. 36 rats were divided into 3 groups (allogeneic dermis, acellular xenogeneic dermis, Vaseline gauze) The labeled P5 cells were transplanted to the wound to observe the shedding time and the number of living cells transplanted. Results Allogeneic skin as the wound cover shed more time than the other two coverings, with a large number of viable cells; while the acellular dermal cover group was superior to the Vaseline gauze cover group. Conclusion The application of allogeneic skin as a wound covering can protect bone marrow stromal cells from transplantation and successfully construct a model of wound skin defect for the study of transplantation of bone marrow stromal stem cells.
