目的:利用基因芯片技术研究寒性中药大黄对2,4-二硝基苯酚致实热证模型大鼠肝脏全基因表达谱的影响,从基因水平分析大黄作用机制。方法:SPF级Wistar大鼠24只,随机分为空白对照组、模型对照组和大黄治疗组。造模后6h取肝组织,应用RatRef-12基因芯片检测各组大鼠肝脏基因表达,筛选差异表达基因,进行基因功能分类。结果:大黄水煎液组与实热模型组比较有177条差异表达基因。其中可注释基因113条,查询到29项显著性基因功能。其中生物学过程24项,主要为代谢过程功能包含59(52.21%)条差异基因;分子功能2项,主要涉及催化活性包含61(53.98%)条差异基因,其中氧化还原酶活性包含26(23%)条差异基因。结论:寒性中药大黄可调节代谢相关的基因降低机体代谢水平,实现其“清热泻火”作用。
OBJECTIVE: To study the effect of cold-blooming rhubarb on the gene expression profile of liver induced by 2,4-dinitrophenol in rats by real-time PCR, and to analyze the mechanism of rhubarb from the gene level. Methods: Twenty-four SPF Wistar rats were randomly divided into blank control group, model control group and rhubarb treatment group. Liver tissues were harvested at 6h after modeling. RatRef-12 gene chip was used to detect the gene expression of liver in each group. The differentially expressed genes were screened for gene function classification. Results: There were 177 differentially expressed genes in Rhubarb decoction group compared with the real model group. Among them, 113 genes were annotated and 29 significant genes were found. Among them, there are 24 biological processes, including 59 (52.21%) differential genes in the metabolic process, 2 molecular functions, mainly involved in the catalytic activity of 61 (53.98%) differential genes, of which oxidoreductase activity includes 26 %) Difference gene. Conclusion: The rhubarb can modulate the metabolism related genes and reduce the body metabolism, and achieve its “heat purging fire” effect.
