作者以γ-射线照射感染伯氏疟原虫的蚊子反复叮咬免疫BALB/c株小鼠。再以1.4×10~8免疫小鼠脾细胞和1.4×10~7浆细胞瘤(P3U1)细胞,按Kohler和Milstein方法杂交。杂交细胞瘤培养物是否产生子孢子抗体,用间接免疫荧光试验或子孢子沉淀(CSP)反应测定。经测定600个杂交细胞瘤培养物,其中5个的上清液于融合后2周有子孢子抗体产生,但只有1个杂交瘤细胞系(3D11)繁殖成功,现保种于含10%胎牛血清
The authors immunized BALB / c strain mice repeatedly with bite of mosquitoes infected with P. berghei by gamma-ray irradiation. The spleen cells and 1.4 × 10 ~ 7 plasma cell tumor cells (P3U1) were immunized with 1.4 × 10 ~ 8 cells and crossed with the method of Kohler and Milstein. Whether hybridoma cultures produce sporozoite antibodies is determined by indirect immunofluorescence assay or sporulation (CSP) reaction. After 600 hybridoma cultures were assayed, five of the supernatants produced sporozoite antibodies 2 weeks after fusion, but only one hybridoma cell line (3D11) was successfully propagated and was seeded in 10% fetal bovine serum
