目的 探讨Oncolyn对温石棉所致肺泡巨噬细胞氧化损伤的影响。方法 应用支气管灌洗的方法获得大鼠肺泡巨噬细胞 (AM) ,纯化后与终浓度 5 0 μg/ml的温石棉分别孵育 2h和 4h ,在孵育的同时加入不同浓度的Oncolyn ,检测AM中DNA损伤状况以及抗氧化酶SOD、GSH Px的活性。结果 肺泡巨噬细胞与温石棉共同培养 2h后 ,DNA损伤和SOD、GSH Px活性增加 ,表现出彗星出现率和DNA迁移长度增加 ;当培养 4h后 ,彗星出现率比培养 2h更高 ,DNA迁移长度不再增加 ,而SOD、GSH Px活性均下降。在AM与温石棉作用的同时加入不同浓度的Oncolyn后 ,DNA损伤减轻 ,抗氧化酶SOD、GSH Px的活性上升。结论 Oncolyn对温石棉所致肺泡巨噬细胞的氧化损伤有一定的缓解作用
Objective To investigate the effect of Oncolyn on oxidative damage of alveolar macrophages induced by chrysotile. Methods Alveolar macrophages (AMs) were obtained by bronchoalveolar lavage. After being purified, chondrocytes were incubated with chrysotile at a concentration of 50 μg / ml for 2 h and 4 h, respectively. At the same time, different concentration of Oncolyn was added to detect AM DNA damage and antioxidant enzymes SOD, GSH Px activity. Results After alveolar macrophages co-cultured with chrysotile asbestos for 2 hours, the DNA damage and the activities of SOD and GSH Px were increased, and the incidence of comet and migration of DNA were increased. After cultured for 4 hours, the incidence of comet was higher than that of 2 hours and the DNA was migrated The length no longer increased, but SOD, GSH Px activity decreased. After adding different concentrations of Oncolyn to AM and chrysotile, the damage of DNA and the activity of antioxidant enzymes SOD and GSH Px increased. Conclusion Oncolyn can alleviate the oxidative damage of alveolar macrophages induced by chrysotile
