目的定量分析甲状腺激素缺乏对围生期大鼠脑GoαmRNA的影响。 方法以PTU溶液灌胃造成大鼠围生期甲减模型 ,部分甲减仔鼠每天腹腔注射T4 2 μg/ 1 0 0 g体重。收集 5、1 5、2 5d的正常、甲减及T4 -替代仔鼠脑组织 ,用竞争性RT -PCR法测定脑GoαmRNA。 结果生后 5、1 5、2 5d的甲减仔鼠脑GoαmRNA水平均较同日龄正常仔鼠明显升高 ,尤以 1 5d时最为显著。T4 -替代可明显减轻这种变化。 结论甲状腺激素对发育期鼠脑Goα基因的表达有下调作用 ,这可能是甲状腺激素调节脑发育的机制之一
Objective To quantitatively analyze the effects of thyroid hormone deficiency on goa mRNA expression in the perinatal rat brain. Methods Perinatal hypothyroidism model was induced by intragastric administration of PTU solution. Some hypothyroid mice were intraperitoneally injected with T4 2 μg / 100 g body weight. Normal, hypothyroidism and T4 - of 5, 15, 25 days were collected to replace the brains of newborn rats, and brain Goα mRNA was determined by competitive RT - PCR. Results Goa mRNA levels in hypothyroid rats at 5, 15, 25 days after birth were significantly higher than those at normal age, especially at 15 days. T4 - Substitution can significantly reduce this change. Conclusions Thyroid hormone has a down-regulation effect on Goα gene expression in developing mouse brain, which may be one of the mechanisms by which thyroid hormone regulates brain development
