目的检测EGF,EGF-R和C-erb B-2在食管癌中的表达,并探讨其相互关系。方法用免疫组化ABC法检测60例食管癌和25例正常食管组织中EGF,EGF-R及C-erb B-2的表达状况。结果 EGF主要定位于细胞膜,EGF-R和C-erb B-2蛋白则主要定位于细胞核,食管癌组织中EGF-R和C-erb B-2表达阳性率分别为58%和18%,明显高于正常对照的24%和0%(P<0.05)。统计分析示EGF-R或C-erb B-2阳性的癌组织具较高的淋巴转移率,EGF与EGF-R双阳者淋巴转移率也较高。结论食管癌中也存在EGF-R的过表达和EGF/EGF-R自分泌循环,EGF-R的高表达对肿瘤淋巴转移有促进作用,预后不良。
Objective To detect the expression of EGF, EGF-R and C-erb B-2 in esophageal cancer and discuss their relationship. Methods The expression of EGF, EGF-R and C-erb B-2 in 60 cases of esophageal cancer and 25 cases of normal esophageal tissues were detected by immunohistochemical ABC method. Results EGF was mainly localized on the cell membrane. EGF-R and C-erb B-2 proteins were mainly localized in the nucleus. The positive expression rates of EGF-R and C-erb B-2 in esophageal cancer tissues were 58% and 18%, respectively. 24% and 0% higher than normal controls (P < 0.05). Statistical analysis showed that EGF-R or C-erb B-2 positive cancer tissues had higher lymphatic metastasis rates, and EGF and EGF-R double positive lymph node metastases also had higher rates. Conclusion EGF-R overexpression and EGF/EGF-R autocrine cycle also exist in esophageal cancer. The high expression of EGF-R promotes tumor lymphatic metastasis and has a poor prognosis.
