目的探讨人血管生成素1(Ang-1)对人胃癌细胞BGC-823基质金属蛋白酶2(MMP-2)表达情况的影响。方法利用腺病毒作为载体将已构建成功的含Ang-1基因的重组腺病毒(Ad-Ang-1)瞬时转染人胃癌细胞BGC-823为实验组,转染编码绿色荧光蛋白的复制缺陷型腺病毒重组体(Ad-GFP)为阴性对照组,未转染的正常细胞为对照组。应用RT-PCR和Western blot印迹检测三组细胞中MMP-2在mRNA和蛋白表达的情况。结果MMP-2在实验组中表达明显高于阴性对照组和空白对照组(P<0.05)。结论Ang-1能显著提高人胃癌细胞BGC-823的MMP-2的表达,提示Ang-1可能具有促进肿瘤细胞侵袭的作用。
Objective To investigate the effect of Ang-1 on the expression of matrix metalloproteinase-2 (MMP-2) in human gastric cancer cell line BGC-823. Methods Ad-Ang-1 recombinant adenovirus containing human Ang-1 gene was transiently transfected into human gastric cancer cell line BGC-823 using adenovirus as a vector and transfected with a replication-defective green fluorescent protein Adenovirus recombinant (Ad-GFP) as a negative control group, untransfected normal cells as a control group. The mRNA and protein expression of MMP-2 in three groups of cells were detected by RT-PCR and Western blotting. Results The expression of MMP-2 in the experimental group was significantly higher than that in the negative control group and the blank control group (P <0.05). Conclusion Ang-1 can significantly increase the expression of MMP-2 in human gastric cancer cell line BGC-823, suggesting that Ang-1 may promote the invasion of tumor cells.
