为提高竹笋短肽得率(TCA-NSI)及水解度(DH),本文对Protamex和Papain同步复合酶解竹笋蛋白制备竹笋短肽的方法进行了系统研究。在单因素试验基础上,进行了四因素五水平的二次正交旋转组合试验,建立了TCA-NSI及DH与各种影响因素的回归模型。结合实际生产,确定了Protamex和Papain酶解竹笋蛋白的最适条件为:复合酶比例1:2、pH值7.0、温度58℃、酶用量9000U/g、酶解时间179min、底物浓度2%。在此条件下,体系TCA-NSI为70.69±0.57%,水解度为25.07±0.71%。
In order to improve the TCA-NSI and DH, we systematically studied the method of simultaneous preparation of bamboo shoot peptides by Protamex and Papain. On the basis of single factor experiment, quadratic orthogonal rotation combination experiment of four factors and five levels was carried out, and a regression model of TCA-NSI and DH with various influencing factors was established. According to the actual production, the optimal conditions for protease of Protamex and Papain were determined as follows: enzyme ratio of 1: 2, pH value of 7.0, temperature of 58 ℃, enzyme dosage of 9000 U / g, hydrolysis time of 179 min, substrate concentration of 2% . Under these conditions, the TCA-NSI of the system was 70.69 ± 0.57% and the degree of hydrolysis was 25.07 ± 0.71%.
