目的探讨水飞蓟宾对肝星状细胞(HSC)增殖的抑制和促凋亡作用。方法取对数生长期的HSC-T6细胞,用完全培养基将细胞悬液的浓度调至1×104个/ml,接种于96孔板,以MTT法分别检测OD值,以流式细胞术检测各组细胞凋亡率。结果随着药物剂量的增加,细胞的增殖受抑,呈明显剂量依赖性。对照组、25umol/L,50 umol/L,100 umol/L,200 umol/L,400 umol/L的凋亡比分别为(5.30±0.97)%、(14.60±1.70)%、(35.40±8.93)%、(47.60±11.34)%、(55.80±15.94)%、(69.60±22.93)%,可见水飞蓟宾剂量依赖性可诱导HSC-T6凋亡。结论水飞蓟宾对HSC增殖和迁移具有明显的抑制作用,其机制可能与其诱导肿瘤细胞的早期凋亡有关,细胞凋亡与细胞周期活动密不可分。
Objective To investigate the inhibitory effect of silybin on the proliferation of hepatic stellate cells (HSC) and its role in promoting apoptosis. Methods The logarithmic growth phase of HSC-T6 cells were treated with complete medium to adjust the concentration of the cell suspension to 1 × 10 4 cells / ml and seeded into 96-well plates. The OD values were determined by MTT assay, respectively. Flow cytometry The apoptosis rate of each group was detected. Results With the increase of drug dose, the proliferation of cells was inhibited in a dose-dependent manner. The apoptotic ratios in the control group were (5.30 ± 0.97)%, (14.60 ± 1.70)%, (35.40 ± 8.93) and ), (47.60 ± 11.34)%, (55.80 ± 15.94)%, (69.60 ± 22.93)%, respectively, showing that silibinin can induce apoptosis of HSC-T6 in a dose-dependent manner. Conclusion Silybin significantly inhibits the proliferation and migration of HSC, which may be related to the induction of early apoptosis of HSC. Apoptosis is closely related to cell cycle activity.
