目的探讨不同强度微波辐射对家兔心脏的损伤作用及其机制。方法分别以强度为50、100、150和200 m W/cm2,频率为2450 MHz微波照射家兔20 min,照射后6 h,取家兔心脏,检测心肌细胞中ATP含量、线粒体呼吸链复合体Ⅳ和Ⅴ活性,HE染色观察心脏组织结构的改变,Western blotting检测心肌细胞内Caspase-3和HSP 70蛋白表达量变化。结果随着微波辐射强度的增加(100、150和200 m W/cm2),辐射组家兔心肌细胞中ATP含量、线粒体呼吸链复合体Ⅳ和Ⅴ活性呈现降低趋势,与对照组相比,差异具有统计学意义(P<0.05)。HE染色结果显示,微波辐射组家兔心肌细胞轻度水肿,肌纤维排列不整,心肌细胞出现明显的损伤。Western blotting结果显示,微波辐射后心肌细胞内Caspase-3和HSP 70蛋白表达水平随着辐射强度的增加呈上升趋势(P<0.05)。结论微波辐射对家兔心脏具有明显的损伤作用,其可能机制是通过改变心肌细胞内应激水平而引发细胞凋亡。
Objective To investigate the injury and its mechanism of rabbit heart under different intensities of microwave radiation. Methods The rabbits were irradiated with microwaves of 50, 100, 150 and 200 mW / cm2 at a frequency of 2450 MHz respectively for 20 min and 6 h after irradiation. The hearts of rabbits were harvested for the determination of ATP content, mitochondrial respiratory chain complex Ⅳ and Ⅴ. The changes of cardiac structure were observed by HE staining. The expression of Caspase-3 and HSP70 protein in myocardium were detected by Western blotting. Results With the increase of microwave radiation intensity (100, 150 and 200 mW / cm2), the ATP content and the activity of Ⅳ and Ⅴ of mitochondrial respiratory chain complex in the irradiated group decreased compared with the control group Statistically significant (P <0.05). The results of HE staining showed that the myocardial cells in the microwave radiation group had mild edema, the muscle fibers were not arranged well, and the myocardial cells were obviously damaged. The results of Western blotting showed that the expression of Caspase-3 and HSP70 in myocardial cells increased with the increase of radiation intensity (P <0.05). Conclusion The microwave irradiation has a significant damage on the heart of rabbits. The possible mechanism is that apoptosis is induced by changing the stress level in cardiomyocytes.
