目的:探讨骨髓间充质干细胞(MSCs)分化为皮肤附属器的可行性。方法:抽取小型香猪的骨髓,经密度梯度离心分离、体外纯化间充质干细胞及培养扩增后,应用5-溴脱氧尿嘧啶(BrdU)标记技术进行细胞标记。将已标记的细胞(1×10~6)以注射方式回植到提供骨髓的猪的皮内及皮下,分别于注射后1、2、4周取材,常规石蜡包埋、切片,行 HE 染色及 BrdU 和角蛋白免疫荧光染色,激光共聚焦显微镜观察。结果:部分 BrdU 阳性标记细胞出现在皮脂腺周围的导管中参与皮脂腺导管的形成,这些细胞经双染证实也同时表达角蛋白。提示这部分双染的导管细胞来自于植入的 MSCs。结论:在皮肤微环境下骨髓间充质干细胞可能分化为皮脂腺导管细胞。
Objective: To investigate the feasibility of differentiation of bone marrow mesenchymal stem cells (MSCs) into skin appendages. Methods: Bone marrow of miniature piglets were isolated and purified by density gradient centrifugation. Mesenchymal stem cells were purified in vitro and cultured. After being cultured and expanded, the cells were labeled with 5-bromodeoxyuridine (BrdU). The labeled cells (1 × 10 ~ 6) were transplanted into the bone marrow-provided pigs intradermally and subcutaneously by injection at 1, 2 and 4 weeks after injection, respectively. The cells were paraffin-embedded, sectioned and stained with HE And BrdU and keratin immunofluorescence staining, laser confocal microscopy. Results: Some BrdU positive labeled cells appeared in the duct around the sebaceous gland and participated in the formation of sebaceous duct. These cells were confirmed by double staining and expressed keratin at the same time. It is suggested that this partially double-stained ductal cell comes from implanted MSCs. CONCLUSION: Bone marrow-derived mesenchymal stem cells may differentiate into sebaceous ductal cells under the skin microenvironment.
