目的探讨前列腺癌(PCa)组织中CD146(Mel-CAM,MUC18)的表达与其微血管形成的关系。方法采用免疫组化(EnVision+TM)方法检测45例PCa、24例良性前列腺增生(BPH)组织中CD146蛋白的表达和CD34标记的肿瘤微血管数目和密度(MVD)值。结果CD146蛋白在BPH和PCa组织阳性表达率分别为12.5%和82.2%;BPH与PCa组织的MVD值分别为20.40±10.25和36.10±9.94;两者均较对照组(BPH)差异有统计学意义(P<0.01)。CD146阳性组与CD146阴性组间的MVD值存在显著性差异(P<0.01)。CD146的表达与PCa分化程度、临床病理分期无统计学意义(P>0.05)。结论CD146表达是PCa形成的早期事件,CD146可能参与了前列腺癌浸润和转移过程;CD146在前列腺癌微血管形成过程中发挥了重要作用;CD146可作为判断前列腺癌预后的指标和治疗的新靶点。
Objective To investigate the relationship between the expression of CD146 (Mel-CAM, MUC18) and its microvessel formation in prostate cancer (PCa). Methods Immunohistochemistry (EnVision + TM) method was used to detect CD146 protein expression and CD34-labeled tumor microvessel density (MVD) in 45 cases of PCa and 24 cases of benign prostatic hyperplasia (BPH). Results The positive rates of CD146 protein in BPH and PCa tissues were 12.5% and 82.2%, respectively. The MVD values of BPH and PCa tissues were 20.40 ± 10.25 and 36.10 ± 9.94, respectively. There was significant difference between the two groups (P <0.01). There was a significant difference in MVD between CD146 positive group and CD146 negative group (P <0.01). CD146 expression and PCa differentiation, clinical pathological stage was not statistically significant (P> 0.05). Conclusion CD146 expression is an early event in PCa formation. CD146 may play an important role in the invasion and metastasis of prostate cancer. CD146 plays an important role in angiogenesis of prostate cancer. CD146 may serve as a new target for prognosis and prognosis of prostate cancer.
