胰岛素样生长因子-Ⅰ在大鼠骨骼肌运动性损伤卫星细胞再生中的作用

目的通过研究胰岛素样生长因子-Ⅰ(IGF-Ⅰ)对骨骼肌运动损伤后卫星细胞再生的影响,探讨IGF-Ⅰ在骨骼肌运动损伤修复中的应用前景。方法使用SD大鼠1周力竭模型,首次力竭后第1、3、5天给予右侧腓肠肌中部的肌膜下分别注射外源性IGF-Ⅰ300ng/ml(实验组)、生理盐水1ml(对照组),分别在首次力竭后7、9、11、17、24、30d取材,进行骨骼肌HE染色光镜观察,电镜超微结构观察和增殖细胞核抗原(PCNA)表达的比较。结果首次力竭后7、11、17d实验组卫星细胞明显增多,对照组胶原纤维增多,30d实验组肌纤维塑形良好,对照组可见瘢痕愈合;电镜观察对照组可见肌浆网扩张,线粒体肿胀,还有凋亡细胞出现;实验组线粒体形态较正常,糖原颗粒增多,可见新生的肌纤维。PCNA指数实验组明显高于对照组(P<0.05)。结论IGF-Ⅰ对运动性骨骼肌损伤后的卫星细胞再生有促进作用。 Objective To investigate the effect of insulin-like growth factor-Ⅰ (IGF-Ⅰ) on satellite cell regeneration after skeletal muscle injury and to explore the potential application of IGF-Ⅰ in the repair of skeletal muscle injury. Methods Exhaustive models of SD rats were established. One week after the first exhaustive exercise, the rats were injected subcutaneously with 300ng / ml IGF-Ⅰ (experimental group), 1ml Control group) were taken at 7, 9, 11, 17, 24 and 30 days after the first exhaustion, respectively. HE staining was used to observe the skeletal muscle ultrastructure and the expression of proliferating cell nuclear antigen (PCNA) was compared. Results The number of satellite cells in the experimental group increased significantly after the first exhaustion, and the number of collagen fibers in the control group increased. In the control group, the muscle fibers were well shaped in the 30th day. The scar was healed in the control group. In the control group, the sarcoplasmic reticulum was dilated, Apoptotic cells also appeared; experimental group mitochondrial morphology than normal glycogen particles increased, visible newborn muscle fibers. PCNA index experimental group was significantly higher than the control group (P <0.05). Conclusion IGF-Ⅰ may promote satellite cell regeneration after exercise-induced skeletal muscle injury.
