目的:建立固相萃取结合高效液相色谱法测定人血浆中酒石酸唑吡坦浓度。方法:血样经固相萃取法提取,采用大连依利特色谱柱(Hypersil BDS C_(18)),流动相:甲醇-乙腈-水(45∶45∶110),流速1.0ml·min~(-1),柱温:35℃;检测波长:254 nm,内标法定量。结果:血浆中酒石酸唑吡坦浓度在0.511 5~4.092 0μg·ml~(-1)线性关系良好,相对回收率大于90%,精密度日间、日内RSD均小于10%。结论:该测定方法简单、快速、准确、干扰小,适用于临床测定酒石酸唑吡坦药物浓度。
Objective: To establish a solid phase extraction coupled with high performance liquid chromatography for the determination of zolpidem tartrate in human plasma. Methods: The blood samples were extracted by solid phase extraction (SPE). The sample was separated on a Dalian Hypersil BDS C 18 column with a mobile phase of methanol - acetonitrile - water (45:45:110) at a flow rate of 1.0 ml · min -1 ), Column temperature: 35 ℃; detection wavelength: 254 nm, internal standard method. Results: The linearity of zolpidem tartrate in plasma ranged from 0.511 5 to 4.092 0 μg · ml -1, and the relative recovery was more than 90%. The RSDs of intra-day and intra-day precision were less than 10%. Conclusion: The method is simple, rapid, accurate and small interference, suitable for clinical determination of zolpidem tartrate drug concentration.
