目的研究类黄酮化合物对前列腺癌DU145细胞表达PSCA的抑制作用。方法采用光学显微计数法,考察染料木黄酮、木犀草素以及槲皮素对DU145细胞的增殖抑制活性;采用荧光定量PCR和Western-blot技术,分析三种类黄酮化合物作用DU145细胞后,PSCA基因mRNA表达和蛋白表达水平的变化。结果三种类黄酮化合物均能抑制DU145细胞增殖,并抑制DU145细胞表达PSCA。结论类黄酮化合物可通过降低PSCA表达抑制DU145细胞增殖。
Objective To study the inhibitory effect of flavonoids on the expression of PSCA in prostate cancer DU145 cells. Methods The inhibitory activity of genistein, luteolin and quercetin on the proliferation of DU145 cells was investigated by optical microscopy. The expression of PSCA gene in DU145 cells was analyzed by fluorescence quantitative PCR and Western-blot. mRNA expression and protein expression level changes. Results All three flavonoids could inhibit the proliferation of DU145 cells and inhibit the expression of PSCA in DU145 cells. Conclusion Flavonoids can inhibit the proliferation of DU145 cells by decreasing PSCA expression.
