目的:观察系统性红斑狼疮(SLE)患者外周血单一核细胞(PBMC)IKB激酶(IKK-β)表达水平,并探讨其临床意义。方法:实时荧光定量聚合酶链反应技术检测IKK-βmRNA表达水平;蛋白印迹法检法检测IKK-β、磷酸化IKK-β表达水平,并观察IKK-β的表达与疾病活动的相关性。结果:(1)SLE患者TNF水平(242.88±49.95pg/ml)显著高于正常对照组(115.23±31.55pg/ml),差异有统计学意义(p<0.05);(2)SLE患者IKK-β基因表达水平(1.61±1.3)与正常人(1.57±1.38)相比无差异(p>0.05);(3)SLE患者磷酸化IKK-β蛋白表达水平(1.55±0.21)显著高于正常人(0.95±1.1),差异有统计学意义(p<0.05);(4)SLE患者IKK-β表达与抗核抗体、dsD水平呈正相关(p<0.05)。结论:SLE患者外周血IKK-β表达明显增高,与SLE发病及病情活动相关,提示IKK-β在SLE的发病中发挥重要的作用。
Objective: To investigate the expression of IKB kinase (IKK-β) in peripheral blood mononuclear cells (PBMC) in patients with systemic lupus erythematosus (SLE) and to investigate its clinical significance. Methods: The expression of IKK-βmRNA was detected by real-time fluorescence quantitative polymerase chain reaction (RT-PCR). The expression of IKK-β and phosphorylated IKK-β was detected by Western blotting. The correlation between IKK-βexpression and disease activity was also observed. Results: (1) TNF levels in SLE patients (242.88 ± 49.95pg / ml) were significantly higher than those in normal controls (115.23 ± 31.55pg / ml) (p <0.05); (2) The level of phosphorylated IKK-βprotein in SLE patients (1.55 ± 0.21) was significantly higher than that in normal subjects (1.61 ± 1.3) and 1.57 ± 1.38 (P> 0.05) (0.95 ± 1.1), the difference was statistically significant (p <0.05). (4) The expression of IKK-β in SLE patients was positively correlated with antinuclear antibody and dsD levels (p <0.05). Conclusion: The expression of IKK-β in peripheral blood of patients with SLE is significantly higher than that of patients with SLE. It suggests that IKK-β plays an important role in the pathogenesis of SLE.
