目的探讨多西紫杉醇对人宫颈癌细胞系Hela的放射增敏作用。方法2004年在哈尔滨医科大学附属肿瘤医院肿瘤研究所采用克隆形成实验分析不同浓度多西紫杉醇以及多西紫杉醇不同预处理时间对Hela细胞生长抑制的影响,用多靶单击拟合模型方程测定各组细胞放射增敏比评价增敏效果;四甲基偶氮唑蓝(MTT)方法检测1.0nmol/L多西紫杉醇对2Gy/次常规分割放疗和1Gy/次、0.5Gy/次低剂量分割放疗方式的增敏效果。结果1.0nmol/L和5.0nmol/L多西紫杉醇放射增敏比分别为1.26和1.69。5.0nmol/L多西紫杉醇分别预处理3h、6h、12h、24h的放射增敏比分别为:1.21、1.31、1.62、1.71,预处理6~12h期间细胞放射增敏比增高速率最快。1.0nmol/L多西紫杉醇对2Gy/次、1Gy/次(×2)、0.5Gy/次(×4)分割放疗组的放射增敏比分别为:2.03、2.51、3.57;多西紫杉醇增敏比分别为:1.41、1.82、2.92,每组中放射增敏比较多西紫杉醇增敏比高。结论多西紫杉醇对宫颈癌细胞系Hela有良好的放射增敏作用,对低剂量分割放射的增敏作用更强。
Objective To investigate the radiosensitization effect of docetaxel on human cervical cancer cell line Hela. Methods In 2004, Harbin Institute of Tumor Hospital Affiliated Cancer Institute cloning formation experiment analysis of different concentrations of docetaxel and docetaxel different pretreatment time Hela cell growth inhibition, measured by a multi-target single-click model equation The radiosensitization ratio of the group of cells was used to evaluate the sensitization effect. MTT assay was used to detect the effect of 1.0nmol / L docetaxel on 2Gy / sub-routine radiotherapy and 1 Gy / second, 0.5 Gy / sub-radiotherapy Sensitive effect of the way. Results The radiosensitivities of 1.0 and 5.0 nmol / L docetaxel were 1.26 and 1.69.5.0 nmol / L pretreatment with docetaxel for 3 h, 6 h, 12 h and 24 h, respectively. The radiosensitivities were 1.21, 1.31,1.62,1.71, pretreatment 6 ~ 12h period, the rate of cell radiosensitization increased fastest. The radiosensitivities of 1.0nmol / L docetaxel to 2Gy / times, 1Gy / times (× 2) and 0.5Gy / times (× 4) radiotherapy groups were 2.03,2.51,3.57, respectively. Ratio were: 1.41,1.82,2.92, radiosensitization in each group more than the high sensitivity of docetaxel. Conclusion Docetaxel has a good radiosensitization effect on cervical cancer cell line Hela, and has stronger sensitizing effect on low dose radiation.
